Associate Professor (A/Prof) Tania Crotti is a Medical Researcher and Education Specialist in the School of Biomedicine (Sch Biomed), Faculty of Health and Medical Sciences (FHMS) at the The University of Adelaide. As the Manager of Postgraduate Coordinators, Sch Biomed and PGC for the Sch Biomed, School of Allied Health and Practice, and the Adelaide Medical School (AMS), A/Prof Crotti works with Higher Degree Research (HDR) candidates across the faculty, while actively supervising Honours and PhD candidates in her fields of research. To support supervisors, PGCs and professional staff working with HDRs, A/Prof Crotti co-leads the HDR Supervision Community of Practice (HDR CoP).
A/Prof. Crotti was awarded a BHealthSci (Hons) in 1997 then worked as a Research Assistant in the Department of Pathology, The University of Adelaide. In 2002 she completed her PhD on "The mechanisms responsible for pathological bone remodelling in inflammatory conditions." (funded by a Dora Lush, NHMRC Scholarship). A/Prof. Crotti subsequently worked as a postdoctoral fellow at the Harvard Affiliated Beth Israel Deaconess Medical Center, Boston USA 2003-2008. She was awarded a CJ Martin, NHMRC Fellowship to characterise the mechanisms regulating gene expression during osteoclast differentiation. In 2006 A/Prof. Crotti was awarded an American Arthritis Foundation Fellowship to investigate the role of cell-substrate interactions and genes associated with terminal osteoclast activation.
A/Prof. Crotti returned to Adelaide in 2008 to continue her work in the field of Osteoimmunology. Her research interests have included peri-implant loosening, periodontal disease and Rheumatoid Arthritis (RA). Current work investigates mechanisms of bone loss and pain associated with chemotherapy and RA. A/Prof Crotti also has a particular interest in the well being of others with work around the patient experience in people with Endometriosis and Juvenile Idiopathic Arthritis, adjunct therapy for Veterans with PTSD and the creation of community to support of Higher Degree Research (HDR) Candidates.
In recognition of A/Prof. Crotti's contributions to undergraduate and postgraduate research training at the University of Adelaide, A/Prof Crotti has received the FHMS Executive Dean’s Learning and Teaching Prize and Development Award (2013), a Beacon Award for Developing Coursework Students as Researchers and using Evidence-Based Practice (2017), Adelaide Medical School (AMS) Higher Degree Research (HDR) Award (2018) and an Executive Deans Award for Teaching Excellence (FHMS) (2022). In 2024 she was appointed the FHMS Representative in the Adelaide Education Academy (AEA) Executive Group and received the Stephen Cole the Elder Awards for Excellence in HDR Supervisory Practice.
For A/Prof Crotti's leadership and mentorship in research education, A/Prof. Crotti has been accredited as a Senior Fellow in Higher Education Advance (SF HEA, 2021). Her contribution to postgraduate education and training has been recognised by the Australian Council of Graduate Research (ACGR) Excellence in Graduate Leadership Award (2023) and she is a current mentor in the Asia Pacific Women in Leadership (APWiL) program (2023/24).

Associate Professor (A/Prof) Crotti leads the Bone and Joint Osteoimmunology Laboratory in projects to understand the cell biology and pathological processes involved inflammation-induced bone loss (Osteoimmunology) such as that seen in rheumatoid arthritis and periodontal disease as well as cancer and chemotherapeutic treatments.

Through ongoing collaborations with scientific and clinical researchers, the laboratory's work takes a holistic and systemic approach when understanding the associated effects of disease and treatment on the musculoskeletal system together with pain and well being using pre-clinical and clinical models. 

With an interest in pain and well being, A/Prof Crotti is also involved in work on the language of pain in Endometriosis and the use of adjunct therapies for veterans with post-traumatic stress disorder (PTSD) using qualitative and linguistics analysis.

As a member of the Adelaide Education Academy Executive Group and contributor to the Adelaide Development Program for Educators and Professionals who Teach (ADEPT) program, A/Prof Crotti is active in evidence-based teaching and research.  A/Prof Crotti has presented internationally on the preparation of undergraduate students for a career in research and published on authentic assessment. Her interests also centre around supporting supervisors and Postgraduate Coordinators (PGCs) in supporting a positive and successful candidature for Higher Degree Research (HDR) candidates.  Studies investigating the importance of community in the HDR experience and placing the HDR student at the centre have been supported by Learning and Teaching Advancement Grants with findings and expertise shared by publications in THE Campus, Times Higher Education presentations at the Quality Postgraduate Research conference and through invited webinars (eg "The establishment of a HDR Supervision CoP" to the  Australia Council of Postgraduate Research members).

Research Project: 

Title: Dietary interventions for protection from chemotherapy-induced bone loss

Project description: Drugs used to treat breast cancer cause damage to normal bone structure, leading to adverse symptoms such as loss of bone mineral density and fractures in survivors. Our group is working with industry to explore different dietary interventions to prevent bone loss following treatment with the chemotherapy drug, methotrexate. This project will use breast cancer rodent cancer models treated with chemotherapy. Research techniques include histological analysis, cell death assays, immunofluorescence, real time PCR and small animal handling.

Research Project: 

Title: Objective assessment of pain in Rheumatoid Arthritis utilising neuroimmunology and biophotonics.

Project description: There are currently no objective tests to diagnose or quantify persistent pain.  Nor are there tools to aid the selection of the best drug treatment for each patient.  Persistent pain and enhanced reactivity to pain, including that experienced in RA, is now appreciated to involve bidirectional communication between the peripheral immune, spinal immune and brain immune systems. This project will extend our work from other persistent pain populations to create a novel diagnostic toolkit that can collectively provide an objective differential measure of musculoskeletal disease and pain experience. Using patient biological specimens and clinical data obtained in collaboration with the Rheumatology Department at the Flinders Clinical Centre we are applying novel hyperspectral imaging to objectively assess pain.

Research Project:

Title: Mitigating the effects of periodontal disease on bone  

Up to 90% of the adult population is affected by some form of chronic periodontitis, and almost half being diagnosed with destructive periodontitis (PD). Thus, effective and sustainable treatments modalities need to be available to reduce the alveolar bone recession and tooth loss seen in this population. Histone deacetylases (HDACs) have been identified as a group of specific targets that effectively reduce bone destruction and inflammation in vitro and in vivo.

Our recent in vivo investigations utilised a systemic oral treatment method that demonstrated the ability of broad-acting HDACi 1179.4b to suppress alveolar bone loss in periodontitis.

Preliminary data in an in vitro study of monocytes/macrophages with the Hydrogel and the novel HDACi 1179.4b (unpublished) show no signs of toxicity.

Components of P. gingivalis such as LPS are recognised as main virulence factors responsible for the pathogenesis of PD. Several studies have theorised that P. gingivalis LPS can activate cells by binding to different membrane receptors expressed on macrophages to induce inflammation as well as bone resorption. Therefore, to simulate a periodontopathic inflammatory response, we will have bone and immune cells exposed to P. gingivatis LPS and treated with the HDACi hydrogel.

Administration of HDACi hydrogel to bone cells (osteoclasts and osteoblasts) and immune cells (monocytes and macrophages) with the presence of P. gingivalis LPS in vitro aims to investigate the efficacy of HDACi 1179.4b hydrogel compound on bone and immune cells in a simulated inflammatory environment to mimic periodontal disease.

Outputs include: RT PCR, immunocytochemical staining, scanning electron microscopy and ELISA.

Research Project :  We hypothesise that the topical local administration in a murine model of periodontitis, in the form of an oral ‘gingival paste’, will be a safe and effective target for the pathological cellular activity, increasing efficacy and reducing the risk of any potential side effects. This will be assessed by microCT analysis of bone, immunohistocytochemistry for inflammatory and bone cell markers and newly applied HDAC Enzyme Immunoassay Kits.
