Dr Mark Gibson - BSc (Hons), PhD, B. Pharm(Hons).
Dr Gibson is an experienced researcher with a long track record in the field of the cell and molecular biology of extracellular matrix. In particular he is studying the biology of elastic fibres, structures responsible for tissue elasticity. In the early part of his career Dr Gibson was responsible for identifying, cloning and sequencing of several of the molecular components of these structures. He has since focussed on the molecular assembly of these complex structures and on the role they play in growth factor regulation. This process becomes aberrant in incurable fibrotic diseases involving major organs such as heart, lung, kidney and liver and genetic diseases such as Marfan syndrome. Currently Dr Gibson's lab is investigating the role of elastic fibre-associated Latent TGF-beta Binding Proteins (LTBPs) in cellular and animal models of fibrosis and fibrotic diseases.

Dr Mark Gibson has been based since 1980 in Pathology at the University of Adelaide, where he has developed a long and distinguished track record on the cell and molecular biology of extracellular matrix, particularly on the difficult area of elastic fibre research.
Major Scientific Achievements
- First identification and characterization of the undegraded form of type VI collagen.
- First identification and characterization of MAGPs-1 and-2 and MP78/70 (ßig-h3).
- First purification of fibrillin-1 from tissue
- First molecular cloning of MAGP-1 and MAGP-2.
- First identification and cloning of fibrillin-like protein (later known as LTBP-2).
- Identification of MAGPs -1 and -2 as regular periodic components of fibrillin-microfibrils.
- Identification of an elastin / collagen VI binding region on MAGP-1.
- Establishment of MP78/70 (ßig-h3) as a widespread matrix protein.
- Determination of distinct microfibrillar expression patterns for MAGP-1 and MAGP-2.
- Characterisation of the human MAGP-2 gene.
- Elucidation of MAGP-2 / cell interactions.
- First characterization of biglycan and decorin interactions with elastic fibre components.
- Establishment of ßig-h3 as a covalent component of collagen VI microfibrils.
- Defining the molecular complexing of ßig-h3 with biglycan and decorin.
- Mapping of MAGP-1 and MAGP-2 binding sites on fibrillins and microfibrils.
- Mapping of LTBP-2 binding sites on fibrillins and microfibrils and establishing its competition for binding with latent TGF-beta complexes.
- Defining LTBP-2 interactions with heparin and heparan sulphates, perlecan and syndecan-4.
- Establishing that LTBP-2 competes with tropoelastin for binding to fibulin-5 and heparan sulphates indicating a negative modulatory role in elastic fibre assembly
- First identification of high-affinity interaction of FGF-2 with LTBP-2, defining the binding site and showing inhibition of FGF-2 activity
- Discovery that LTBP-2 elevates TGF-beta expression in several cell types and identification of the domain responsible for this activity.
- Identification of signalling pathways involved in LTBP-2 elevation of TGF-beta expression
- Discovery that LTBP-2 is elevated in skin fibrosis and co-localises with FGF-2 in fibrotic lesions
